PT-141 is a synthetic melanocortin receptor agonist that primarily activates MC3R and MC4R in the hypothalamus. It works centrally in the brain to stimulate sexual arousal by increasing dopamine release in the medial preoptic area, initiating neural signals for sexual response, and can trigger arousal without direct sexual stimulation. Unlike PDE5 inhibitors, it does not rely on nitric oxide pathways.
